In this photo, the arrow points to the reaction tube floating in the waterbath. The waterbath temperature is monitored by the yellow thermometer, and kept at the optimum temperature for the enzyme being used (usually, but not always, 37ºC). Often, 1 hour is sufficient for digestion, but longer times are frequently used, such as overnight, to assure complete digestion by the enzyme.
