The correct answer for question when the sequence of bases has one base change which would mean "One amino acid will be replaced with a different amino acid", says "The mutation would most likely result in a non-functional protein", but I thought that wasnt necessarily true because as we learned on occasions one amino acid will not effect the function.

The DNA sequence in the question is given to you 5'-3'. When this template is transcribed into mRNA, it will have to be read in the 3'-5' direction in order to be making RNA in the 5'-3' direction. So you have to read this strand from right to left.
