The notes said that the melting temperature of DNA is affected by GC concentration, but does AT concentration affect it also? Why or why not?


The concentrations cannot be separated from each other. If you increase the GC concentration, you are of course decreasing the AT concentration, and vice versa. The real point is that if you make the strands easier to separate, you will lower the melting temperature.
