In DNA replication, is helicase used in both eukaryotes and prokaryotes? What about in transcription?

Yes, although the specific names of the molecules will vary depending on the process and the organism, there is a helicase activity in both prokaryotes and eukaryotes, in both replication and transcription. Any time you have a need to unwind the ds-DNA, you'll need a helicase activity to do that.
