What exactly is the purpose of nuclease in DNA compaction in eukaryotes? How does removing the phosphodiester linkage between nucleic acids make a strand of DNA more compacted?

This is also addressed in another question, but bears repeating. The slide to which you are referring described an experiment conducted to study the organization of chromatin. This is not something a cell is doing on its own. Quite the contrary, the effect of introducing nucleases into the nucleus where your genome is would be devastating, and almost certainly lethal.
