For sickle cell mutation to occur, does the Glu have to change to Val in one or in both beta chains of the hemoglobin?


This is the difference between being a carrier for the disease and having the full-blown disorder. A carrier has one wild-type gene and one mutated gene for beta globin, and therefore is making some good beta-globin and some mutated. But for any given molecule of hemoglobin, both of its beta chains need to be wild-type.
