I am unclear on how we check if a CELL has a VECTOR (Selection Step). I understand the Screening step where we can see if a vector has an insert, but not the Selection step.

Transformation has a fairly low probability of success. So most of the cells that make it through an attempted transformation will NOT have any vectors get into them. These will get in our way and confuse our results later, so we kill off any cell that does not at least take in a vector. This is almost always done by an antibiotic resistance gene on the plasmid that won't be found in the host's genome.
