Do the transport vesicles, when reaching their destination, simply come off the MT and motor proteins?

Yes and no. Yes, the motor proteins can release from the MT once they reach their destination, but no, the motor proteins are part of the vesicle membrane, so they don't "let go" of the vesicle.
