Can you get the C terminus into the cytosol without an internal signal sequence?

Yes, for example if you have an amino terminal signal peptide and a stop transfer sequence, your N-terminus would be in the lumen of the ER, your C-terminus would be in the cytoplasm, and you would cross the membrane exactly one time without needing an internal signal peptide.
