Is it safe to say that if a protein's signal sequence is at the N-terminus it will be chopped off and result in the N-terminus of that protein being in the lumen? Once the signal peptide is chopped off, does this mean the signal peptide cannot become a membrane spanning region?

Yes, that's what that means. Nicely said. :)
