For molecules that undergo passive diffusion, do they require an NES or NLS as well?


They would not be required to have one, but it's possible for it to occur. Think about the localization of proteins that can freely pass across the NPC. In a short time, half the molecules would be in the nucleus and half in the cytoplasm. But if you need all of them to be in, for example, the nucleus, you'd need to give it an NLS so that if one of them "wandered" into the cytoplasm it would immediately be shepherded back into the nucleus where it "belongs."
