Does proofreading always happen? And if it does not, can mismatch repair enzymes still do their job if proofreading did not happen before or can mismatch repair only happen after proofreading? / If mismatch repair is happening, does that automatically tell you that proofreading has already happened and the error frequency has already been decreased by 100 and will then be decreased by 100 again with the mismatch repair enzymes?


Proofreading isn't an "occasional" procedure. DNA polymerases either proofread each base they add or they can't do proofreading at all. Granted, they don't always work, but that doesn't mean they don't always try!
