I was wondering how and why do the enzymes responsible for 5' modification of eukaryotic mRNA and DNA Ligase create phosphodiester linkages?


Both of these reactions require the energy of an ATP hydrolysis. Ligase creates phosphodiester bonds in just the same way as we learned in the DNA replication process discussed earlier in this class. The 5' capping enzyme complex creates a 5' to 5' bond in which there are three phosphates bridging the two adjacent nucleotides.
