If an in vitro experimental system omits something that is critical to a specific process like transcription or translation, then one can assume that that "something" is not present in vitro?


Yes, in an artificial environment of your (the researcher's) creation, if you didn't put it in the tube, it's not there. If you're trying to create an in vitro DNA replication system, and you put ss-DNA in the tube with DNA polymerase III and dNTPs, you shouldn't expect anything to happen, because you left out primase and NTPs.
