Methods: Plants (2 weeks old) were grown on sealed 0.5 x MS agar (3% sucrose) plates. For the observation of water loss, the lid was removed and plants were exposed to open air. Pictures were taken in 1 min intervals for 2 hours. Following desiccation, the plate was sealed again and the plants began to recover. Pictures were taken in two-minute intervals for one hour during the recovery process. Animation was generated via Animation Shop 3, based on these pictures. 10 T2 PIP1b RNAi plants from 5 lines, together with two wild type plants, were used in this experiment. Below is a picture to show the lines (2 plants/line) derived from independent primary transformants.
