Lecture Activity: DNA Sequencing
An unknown segment of DNA consisting of 12 base pairs was prepared for sequencing using dideoxy fluorescent nucleotides ("bogus" nucleotides). The process, similar to PCR, uses a DNA polymerase and DNA bases to make many copies of fragments of the original DNA molecule.
Fluorescent dideoxy bases mixed with normal DNA bases in the copying mixture are randomly included, cutting short the copying process and producing DNA fragments of varying length, each ending with a fluorescent base.
The folowing COMPLEMENTARY fragments were produced terminating with the colored, fluorescent dideoxy bases.
"-O-" represents the unknown nucleotides in each fragment.
